GRANULATION
Granulation is a key processing step in the production of many solid dosage drugs (OSD). Low-viscosity HPMC (Hydroxypropyl methylcellulose or hypromellose) grades are effective as binders for granulation, which can afford the powders with good flowability and improved compressibility.
PHARMACOAT® and TYLOPUR® are a film coating polymer and binder based on hydroxypropyl methylcellulose. They are particularly suitable for fine granules and granules used in direct compression as highly stable binders which do not interact with active substances. The fine particle size allows good admixture with the vehicle (for instance, active substance, lactose, or cornstarch). They are effective for wet granulation done by fluid bed and high shear mixer as well as for twin screw granulation. They are available in various viscosity grades.
DIRECT COMPRESSION
The manufacturing of tablets by direct compression requires smart excipients to improve flowability and compressibility.
L-HPC is a functional excipient, acting as a dry binder and showing excellent anti-capping properties.
Depending on the final application, several grades are available. For direct compression, LH-21 is the primary grade to test. The highly fibrous LH-11 grade is the most effective for capping issues, whereas NBD-022 is indicated for fast disintegration.
SUSTAINED RELEASE
For sustained release formulations, hydrophilic matrix systems are the simplest technology for oral dosage form as they consist of water-soluble highly viscous polymers being cost effective, robust and easy to prepare.
METOLOSE® SR/ TYLOPUR® SR feature optimized viscosity, hydroxypropoxy content, and particle size to guarantee highly reproducible dissolution profiles. They can prevent bursting and disintegration of the tablets which are leading to an undesired too fast release of the active substance. Moreover, they allow formulators to get the desired prolonged release profile by selecting appropriate viscosity or varying the polymer content in the formulation.
ENTERIC RELEASE
Different solutions for enteric coatings can be chosen depending on the API characteristics.
Shin-Etsu AQOAT® (Hypromellose Acetate Succinate) is an enteric coating material developed for aqueous coatings. Its grades show different patterns of the pH dependency in drug release, which allows targeting specific gastro-intestinal sites for drugs release. HPMCP is an enteric coating agent suitable for solvent based systems. It is dissolvable in various organic solvents.
Shin-Etsu AQOAT® and HPMCP show good stability of the coating layer due to less hydroscopicity.
