MONTANIDE adjuvant for antibodies production

The alternative to Freund’s adjuvants

SEPPIC has combined science, expertise and service in order to develop vaccine adjuvants. These adjuvants, registered under the trademark of MONTANIDE have been optimized in order to improve efficacy and stability of vaccine formulations and to reduce side effects. They are widely registered in commercial vaccines for various target species (pigs, cattle, poultry, fishes ...).

For antibodies production, we have selected a powerful adjuvant, MONTANIDE ISA 50 V2, proved to be as reliable as Freund’s adjuvant with:

MONTANIDE ISA 50 V2 is a mineral oil based adjuvant that has been developed for the manufacture of water-in-oil (W/O) emulsions. It comprises an injectable pharmaceutical mineral oil and an extremely refined emulsifier obtained from mannitol and purified oleic acid of vegetable origin.

New vaccine manufacturing process

MONTANIDE ISA 50 V2 is used at a ratio of 50/50 (volume). SEPPIC takes pride in its new and simple protocol of good emulsion manufacture: the T-connector emulsifying protocol.


Step 1- Prepare one part of MONTANIDE ISA 50 V2 in a vial. Then add one part of antigen aqueous solution.

Step 2- Pump both liquids using a sterile syringe with a needle. Remove the air and discard the needle.

Step 3- Connect the syringe to the T-connector* as shown. Do the same with an empty syringe.


Emulsifying:

  • 20 cycles** at low speed (40 seconds)
  • 80 cycles at high speed (80 seconds)

Step 4- Disconnect the T-connector and add a needle to the syringe. It is ready to inject.

*T-connector: Braun Discofix-3 (Ref: 4095111)
**a cycle: the passing of the formulation from one syringe to the other and back
